Summary:碩士 === 國立陽明大學 === 生醫光電工程研究所 === 98 === We set up a multi-channel fluorescence system that can simultaneously detect the fluorescence signal and measuring the brain of fluorescence signal in vivo. First , in order to established the accuracy and stability in our system. we used the NADH fluorescence in solution which the intensity and spectrum position clear and definite. The NADH fluorescence emission about at 470 nm[1]. Then , we also detected the solution of different concentrations and found the fluorescence intensity increased with the concentration levels. The NADH endogenous fluorescence was measured in suspension of cells. The spectrum of cells was similar to that of NADH solution. In addition to the endogenous fluorescence, we also detected the photosensitize of photodynamic therapy 1 mM ALA treat cells for 4 hrs then measured the PpIX emission spectrum at 637 nm[2]. The other exogenous fluorescence is 5 μΜ Fluo3/AM treat cells for 30 mines and measured the calcium fluorescence spectrum at 530 nm[3-4]. Finally , we used the ischemic stroke for in vivo experimental model to measure the brain endogenous fluorescence signal. In the study[5], we know the NADH level is higher when O2 deprivation. Based on this theory to survey the availability and accuracy of our system. Then the experiment result showed that the NADH fluorescence signal of ischemic brain is higher than normal brain. For these reasons, we can detected the experiment in vitro, ex vivo and in vivo of our system, then it will be a potential and development of subsequent biological studies.
