The performances of milk production in association with SNP genotypes were also investigated. RPL31 gene was the differentially expression gene of milk production performance traits in Taiwan, which was discovered from the previous gene chips study in our laboratory. In foreign country, the SCD1, FASN, ATP1A1 and LEP genes had been proved that not only existed SNPs but also associated with milk production. Eighty two DNA samples of Holstein cattle in southern Taiwan were employed for this study. RPL31 gene was analyzed by SSCP (single strand conformation polymorphism), and found out different patterns. A SNP was found from different patterns of SSCP by sequencing. The gene was genotyped by minisequencing. On the other hand, SCD1, FASN, ATP1A1 and LEP were genotyped by PCR-RFLP (polymerase chain reaction-restriction fragment length polymorphism). Finally, the correlation between genotypes and milk yield, fat yield, fat content and protein content were analyzed. The results showed that the milk yield and fat yield in GG genotype were higher than AA genotype (P < 0.05) in LEP gene.
