Phylogenetic relationships of Dasyatidae in the waters off southwestern Taiwan inferred from mtDNA COI and 12S rRNA gene sequences

碩士 === 國立高雄海洋科技大學 === 漁業生產與管理研究所 === 99 === Dasyatidae is included in Kingdom Animal、Phylum Chordata、Class Chondrichthyes、Elasmobranchii、Myliobatiformes in taxonomy. The part COI and entire 12S rRNA gene sequences in mtDNA were used to analyze phylogenetic relationships of Dasyatidae. A total of 15 specimens were collected from WuChi；BuDai；Keziliao and DongGang respectively from October 2010 to January 2011.653 base pairs are observed in the part COI gene segment﹐and 946 base pairs are observed in the entire 12S rRNA gene segment. Five species include Dasyatis akajei﹐Dasyatis navarrae﹐Neotrygon kuhlii﹐Himantura uarnak and Dasyatis zagei were analyzed and Gymnura japonica was used as outgroup. Neighboor-joining、Maximum parsimony and MrBayes methods were used to construct phylogenetic relationships among these five species. The phylogenetic tree structured respectively by sequences of COI﹐12S rRNA and the link of COI and 12S rRNA can discriminate Himantura uarnak﹐Neotrygon kuhlii and Dasyatis zagei﹐but Dasyatis akajei forms a group with Dasyatis navarrae. The NJ、MP and MrBayes phylogenetic trees structured by sequences of the link of COI and 12S rRNA all indicated that Dasyatis akajei forms a group with Dasyatis navarrae and has the closest relationship with Dasyatis zagei﹐the second closest relationship with Neotrygon kuhli and the third closest relationship with Himantura uarnak .
