PBDEs may leach into the environment when the products are manufactured, used or disposed. PBDEs are persistent and bioaccumulative compounds and have been found not only in the environment, but also in blood of pregnant women and breast milk. Objective: Effects of gestational and lactational exposure to BDE-209 on development and reproduction in F2 male mice offspring. Methods: The pregnant ICR mice were gavaged with corn oil and 500 mg/kg/day BDE-209 in corn oil per day between gestational day (GD) 0 to postnatal day (PND) 21. The doses of BDE-209 administered of the oral reference dose for 1000-fold. The first generation (F1) male rats were mated with F1 generation females from different litters on postnatal day (PND)70. Male adult second generation (F2) offspring were divided to four groups as follows: CC group (control F1 fathers + control F1 mothers), CE group (control F1 fathers + exposed F1 mothers), EC group (exposed F1 fathers + control F1 mothers), EE group (exposed F1 fathers + exposed F1 mothers). F2 offspring were sacrificed at PND 70 to evaluate reproductive functions including: (1) Developmental landmarks: number of birth, male sex rate, pinnae detachment, body hair fuzz appearance, incisor eruption, eye opening, ear opening. (2) Reproductive function: sperm count, sperm motility, chromatin DNA structure, sperm mitochondrial membrane potential, reactive oxygen species generation, serum sex hormone level, testicular DNA content and testicular histopathology. Results: Gestational and lactational exposed to BDE-209 were no significant on the number of birth, male sex rate, developmental landmarks, AGD. There was no significant on organ weight. In reproductive function, a increase of chromatin DNA structure in EE group was observed (P<0.05). The sperm mitochondrial membrane (MMP) was significant decreased in EE group (P<0.05). Furthermore, the percentage of round spermatids was significant decreased in EE group (P<0.05) and the percentage of s-phase was significant increased in EE group than in CC group (P<0.05). Moreover, abnormalities of sperm morphology and testicular histopathology were also significant increase in EE group than CC, CE, EC groups (P<0.05). This study demonstrated that BDE-209 might effect in second generation (F2), we can aim the effect assessment of reproductive of third generation and verify the human health risks associated with exposure to this in the future.
