Notch receptor activation is involved in repression of neurogenesis, maintenance of neural progenitor character, promotion of gliogenesis and drives binary fate choices of neuronal and glial cell. Recent studies show that Deltex is a component of the Notch signaling network that physically interacts with the ankyrin repeats of Notch. However it’s function still unknown. Here, we isolated a zebrafish homologue, deltex1, and showed this gene is specifically expressed in the developing nervous system. We found that overexpression of deltex1 mRNA resulted in downregulation of neural progenitor and proneural markers, and upregulation of post-mitotic neuronal markers before neuronal differentiation, indicating that deltex1 is sufficient to promotes the neuron formation. In addition, markers of glial precursors were down-regulated in deltex1 overexpression embryos, and mature glial cells were upregulated before glial differentiation, suggesting deltex1 is sufficient to promote glial cell formation. In addition, to determine whether expression of deltex1 depends on endogenous Notch signaling, we used mind bomb (mib) embryos characterized with a mutation in the gene encoding a ubiquitin ligase for Delta and display a strong reduction in Notch signaling. The results show that deltex1 is an effector of Delta-Notch pathway. Collectively, our results suggest that deltex1 is involved in vertebrate Delta-Notch pathway and regulation of neurogenesis and gliogenesis.
