Based on its size and taste, the rotifers such as Brachianus plicatilis has been breeding for sea fish fry hatch, as the primary live bait. Rotifer culture, though not difficult, but the cost would be high if the high-density rotifer production with pathogen free demanded, so the willingness of current aquaculture industry to accept the fancy way is limited. Up to date, the cultivation of rotifer is still employ the open-style fish pond in Taiwan, and the high risk in disease spread would be taken. The main purpose of this study is the attempt to strike a compromise between the high price of original pathogen free cultivation and the acceptable price of traditional rotifer cultivation with high pathogen contamination concern. The bag made by cellulose acetate was used as a substrate , filling rotifers and appropriate blend of algae bacteria and then placed the traditional open-style water bath for rotifer cultivation. It was found that cellulose acetate bags with good physical tension, able to withstand cultivating handling and operational needs. Cellulose acetate bags also have a remarkable shielding effect of pathogenic bacteria. The bag with high permeability of ammonia and nitrite would allow the re-movement of these metabolite wasted become effective. The low permeability of oxygen of cellulose acetate bags was demonstrated, by chemical treatment, the efficiency of oxygen was improved by chemical pre-treatment of cellulose acetate bag before usage, indicating oxygen is an important factor at high density rotifer culture in final population, and it might be the important limited factor.
