Excised apical buds were cryopreserved by vitrification, and investigated for the effect of treatmet of different precultures with sucrose medium. And then investigate the effect of different precultures by physiological analysis. The goal of this study is to find out the most optimal protocols of cryopreservation of Aquilaria agallocha Roxb.. After preculture, 1-2 mm apical buds were excised from 80-90 days old plant, and then treated with different duration of PVS2, LS solution and different precultures to find out the most optimal protocols. The results demonstrated that the apical buds precultured on 0.2M sucrose medium for 7 days, and then treated with LS for 60 min at room temperature and PVS2 for 90 min at 0℃. Finally, the survival ratio will be 51.82%. Physiological analysis revealed that preculture treatment decresed relative water content, water potential and osmotic potential, and increased the content of soluble protein and sugar. This result indicated that when the plantlets of Aquilaria agallocha Roxb. underwent stress, plantles could increase drought-enduring and anti-freeze abilities, and thus enhanced the survival ratio. Conclusion that the most optimal protocols of cryopreservation of Aquilaria agallocha Roxb.. was 80-90 day old plants precultured on 0.2M surcrose medium for7 days.
