Phalaenopsis plantlets were transferred to commercial flask and treated with various duration of subculture and acclimatization, and to evaluate the deflask plantlets growth on mixed with sphagnum and bamboo charcoal. The plantlets were cultured on 1/2 MS and Hyponex medium which from flower stalk of Phal. Fortunne Saltzman ‘256’ and Dtps. Queen Beer ‘101’, and only Dtps. Jiubao Red Rose ‘280’ cultured on 1/2MS medium. All the culture medium was supplemented 0.5 mg•L-1 NAA with 0.1 mg•L-1 BA. And the density were 11, 13, 15, 17 and 19 plantlets per flask. After 16-week culture, leaf number of Phal. Fortunne Saltzman ‘256’ and Dtps. Queen Beer ‘101’ cultured on 1/2MS medium is more than Hyponex medium, and the better root number of Phal. Fortunne Saltzman ‘256’and Dtps. Queen Beer ‘101’ are 5.09 and 4.33 respectively, at lower culture density of 11, 13 plantlets per flask. The best root length and dry weight of Dtps. Queen Beer ‘101’ are 2.23 cm and 0.17 g respectively, was also found at lower density. The leaf number and root number will increase when the culture density is decreasing. When plantlets culture with lower and higher culture density of 17,19 plantlets per flask , the average root numbers and leaf number are 2.9, 2.19 and 3.42, 2.83, respectively. The suggestion for increasing plant quality and rooting number is culture on 1/2MS medium with 13 plantlets per flask. Phalaenopsis plantlets were transferred to commercial flask and plant in different culture (10, 12, 14, 16, 18 weeks) with different acclimatization weeks (1, 2, 3 weeks) then cultured in sphagnum. After cultured in sphagnum, the upper part of growth plantlets of traits increase with commercial flask culture time, and had the most root number on commercial flask 14 weeks with 2 acclimatization weeks. The best response on Phal. Fortunne Saltzman ‘256’ was found at acclimatization 2 weeks after 14 weeks of culture, on Dtps. Queen Beer ‘101’ was found at acclimatization 2 and 1 weeks after 16 and 18 weeks of culture, and on Dtps. Jiubao Red Rose ‘280’ was found at acclimatization 1 week after 14 weeks of culture. Phalaenopsis plantlets were transferred to commercial flask and cultured 16 weeks with acclimatization 3 weeks then cultured in bamboo charcoal mixed with sphagnum moss ex vitro for 16 weeks. After cultured in bamboo charcoal mixed (0%, 10%, 20%, and 40%) with sphagnum moss, the best response on Phal. Fortunne Saltzman ‘256’ was found at apply 0%, 10% bamboo charcoal in sphagnum moss, it had the most leaf number, leaf span, leaf length and leaf width. And after four month, the best plantlets growth of traits at mixed 10% bamboo charcoal in sphagnum moss, the survival rate decreased with higher bamboo charcoal percent, and had yellowing root.
