The rate of viable seeds (seed containing embryo) of Paph. grown in the greenhouse at constant temperature 23 ± 1 ℃ and average temperature in the summer / winter 28 ± 2 ℃ / 19 ± 2 ℃ were 19.51 % and 45.70 % respectively. However, the rate of viable seeds of Paph. (Hsinying Web × superbiens var curtisii), Paph. Ho Chi Minh (glaucophyllum × delenatii), Paph. (Maiden Paradise × Gold Acers) ×(Over The Meadow × Over The Mountain), Paph. (Gael × Maudiae), and Paph. Hsinying Web × (Flame Heart × Maudiae) grown in the greenhouse at constant temperature 23 ± 1 ℃ were higher than those of average temperature in the summer / winter 28 ± 2 ℃ / 19 ± 2 ℃. Selfing Paph. In Charm Silver Bell, Paph. (Hsinying Web × superbiens var curtisii), Paph. (Maiden Paradise × Gold Acers) ×(Over The Meadow × Over The Mountain), Paph. henryanum ‘Bear’ Paph. Spicerianum, and Paph. (In-Charm Greenery × spicerianum) were made. Capsules were harvested 210~240 days after pollination. Seed germination rate in the 1/4 MS was better than those of in 1/2MS and Hyponex#1 medium. No seed germination was found in the Hyponex#2 medium. Two phase medium would increase the seed germination rate for Paph. (In-Charm Greenery × spicerianum) and Paph. (Chiara × philippinense alba), especially in the solid medium supplemented with 4 ml aseptic water. Seeds of Paph. (Vintage Nasvest × Learu Hest), Paph. (Lovely Yard × Lovely Land) × Paph. (Winston Churchill × Kimura Promise), Paph. (Mem. Dr. Tsuchiya × Jolly Meadows), and Paph. (In-Charm Greenery × spicerianum) cultured in the Hyponex#1 with activated charcoal would be beneficial for seed germination. However, for Paph. (Space Flight × Hsinying Web), Paph. Hsinying Web × (Flame Heart × Maudiae) medium without activated charcoal would be higher seed germination. Stem section derived from in vitro seedlings of Paph. (Cyberspace × Rudy Leopard) and Paph. (Rudy Leopard × Cyberspace) were used as explants for shoot multiplication. The results showed that Hyponex#2 medium supplemented with peptone 2 g/L, banana 25 g/L, potato 25 g/L, BA 5 mg/L, and NAA 1 mg/L is the better media for shoot multiplication (2.94 shoots / explants ) than 1/2 MS (1.13 shoots / explants). 沈再木 學位論文 ; thesis 106 zh-TW |
