Cell therapy as a new therapeutic alternative for aging skin has come into forefront of medicine. In the previous studies, granulocyte colony stimulating factor-mobilized peripheral blood stem cells (GCSF-mobilized PBSCs) have been the most clinically feasible stem cell source for the majority of cell therapy applications. However, there are few reports on the anti-aging effects of it in aging skin. Therefore, the anti-aging potential of GCSF-mobilized PBSCs for aged skin is worth investigation. This study aimed to investigate whether GCSF-mobilized PBSCs could rejuvenate aged skin by the Lanyu pig model. The male Lanyu pig was injected with GCSF to mobilize PBSCs. Thus, GCSF-mobilized PBSCs collected by leukapheresis increased to 34.2%. The female Lanyu pig was then intradermally transplanted with the collected PBSCs from the male donor to investigate the rejuvenation of aged facial skin. The major survival distribution area of PBSCs was in dermis as shown by chromatin stain and fluorescent in situ hybridization. The results also showed that the transplantation of PBSCs in aged facial skin reduced skin wrinkles and pigmentation, improved skin elasticity and tightening, and promoted epidermal cell renewal based on immunhistochemical analysis.
