A marine-derived, gram-negative γ-proteobacteria, Vibrio sp. C1-TDSG02-1, was isolated from Siaogang Harbor, Taitung, eastern Taiwan for its ability to produce bioactive metabolites. This red-pigmented antibiotic was identified as prodigiosin, which has anti-bacterial, anti-fungal and anti-tumor activities. To increase the production of prodigiosin, the growth of C1-TDSG02-1 was studied under different growth media and cultivation conditions. Bacterial growth is measured by absorbance at 600 nm. A paper-disc agar-diffusion assay was used to quantitatively measure the antimicrobial activity of the antibiotic agent against indictor bacterium Staphylococcus aureus. The concentration of prodigiosin was determined spectrophotometrically by measuring the absorbance value at 535 nm. Among the media tested, medium with nutrient broth supplemented with 1% starch and 0.5% glycerol as carbon source, as well as 1% glycine, and 0.2% tryptone as nitrogen source, favored the production of prodigiosin at 30°C in shake flask fermentation. The yield of prodigiosin in the optimal medium was 483 mg/L for fed-batch fermentation after four days of cultivation in a 10-L stirred tank fermentor, which was 1.12 times higher than by shake flask, with highest prodigiosin production after three days of cultivation.
