Based on the method described by Lunt et al. (1999), four microsatellite markers, A1-1, A1-6, A1-8 and A1-10 were identified. Three alleles in the both A1-1 & A1-10, eight alleles in A1-8, and ten alleles in A1-6 were identified in these studied groups. Heteroduplex DNA electrophoresis was then used to determine different allele of individual microsatellite locus for each sample. AMOVA test, the estimation of the widely used F-statistics, was performed with GenAlex 6.41software based on the allelic polymorphism . It showed that there are significantly different among these groups. Furthermore, genetic distance among these groups was calculated and was used to construct the phylogenetic tree using MEGA 5 software. The tree showed that E. fuscogattus is the most distant from other four groups. E. lanceolatus and E. malabaricus form a clade which is then joined by green grouper to form another clade. This clade was then joined by E. coioides . This result suggest that green grouper is either more closely related to the E. lanceolatus / malabaricus lineage or it is the hybrid between E. lanceolatus and E. coioides. The method described here may help to access the genetic distance of special species in the fish farm, to prevent fish inbreeding, and to increase fish fecundity.
