Current studies suggest that Ganoderma lucidum polysaccharides have a variety of biological functions such as antioxidation, antitumor and immunomodulatory. In this study, we have investigated the effects of mycelia formula (MF) on antioxidative status and anti-aging in senescence accelerated prone mice (SAMP8). 3-month old male and female SAMP8 mice were divided into control and three experimental groups. Experimental groups were supplemented with MF 308 (low dose group), 615 (medium dose group) and 1230mg/kgBW/day (high dose group) for 12 weeks. Food intake and body weights were recorded every week. Open field test, grading score, single-trail passive avoidance test and active shuttle avoidance test were estimated in 11th and 12th week. The effect was evaluated by estimating the activities of lipid peroxidation-malondialdehyde (MDA), oxidation protein products-protein carbonyls (PC), superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) in the liver. In addition, the brain 8-hydroxydeoxyguanosine (8-OHdG), hippocampal and cortex inducible nitric oxide synthase (iNOS) level were assessed to study the effect of MF on brain oxidative DNA damage. The β-amyloid protein of brain was observed. The experimental results showed that the food intake, body weights and open field test were not significant. The total score in grading of the medium and high dose group of male and female SAMP8 mice were significantly lower than the control group (p<0.05). Learning and memory ability of experimental groups were better than the control group (p<0.05). The activities of the antioxidant enzymes such as SOD amd GPx were lowered, but MDA and PC were higher in liver of the control group. The treatment of MF for 12 weeks significantly increased the activities of antioxidant enzyme, decreased the levels of lipid peroxidation and oxidation protein products in the liver. Administration of MF significantly decreased the level of 8-OHdG, iNOS and β-amyloid protein deposition in the brain (p<0.05). In summary, the supplement of MF could reduce the MDA and PC level, increase SOD and GPx activity in the liver, reduce the brain of iNOS, 8-OHdG and β-amyloid protein deposition, and then improve the learning and memory ability. Therefore MF may have the effect of anti-aging.
