The tumor's internal mechanism is very complex, including cancer cells and many different types of stromal cells, immune cells, fibroblasts and endothelial cells,it is often studied by people. We are interesting in the relationship between fibroblasts and cancer. In recent years, from some academic literatures, the organization action will also change the intensity of cell-cell interactions. Therefore, our purpose is to simulate the lung tumor environment, discussion of lung cancer cells in tumor activities.Calculated by the rate of cancer cell proliferation and migration speed, analysis the relationship of stromal cells (fibroblasts) and tissue tensile force impact on cancer cells. The traditional dish is not easy to operate on the cell cyclic stretch (CTS) experiment, and it is easy to die in the long time observation, therefore, we use poly-dimethlsioxane (PDMS) as the material. Experimental condition is designed to meet the chip structure,there are two parts of chip: components and thin film, to combination by oxygen plasma technology. Cells were cultured in the upper component, the use of syringe pump is operating conditions medium flow direction and the performance record of cell, by changing the volume change of the lower component, we can operate the hydraulic switch and cyclic tensile stretch. Measured fibroblasts ??smooth muscle actin fluorescent, to explain fibroblast activation under different experimental conditions, and analysis of cell movement and proliferation number in 24 hours, description activated fibroblasts influence the performance of cancer. Comprehensive experimental results, fibroblast condition medium will increase the rate of cell proliferation and the performance of mobile, If fibroblast is effected by both cancer cells and the role of cyclic tensile, fibroblasts ??SMA fluorescence will be higher than the values of other experimental conditions, and in this condition, fibroblast condition medium for cell proliferation rate and rate of movement performance are higher than other experimental conditions, so in the tumor environment, fibroblasts under the influence of cell-tissue interaction will increase the proliferative capacity and the metastasis performance of cancer cells.
