Summary:碩士 === 逢甲大學 === 綠色能源科技碩士學位學程 === 101 === In this study, the strategy by combining strain improvement and fermentation technology was adopted to improve the ability of Escherichia coli tolerate ethanol, furfural and lignocellulosic hydrolyzate. The recombinant E. coli strain was continuously cultured in the furfural for containing medium more than 60 subcultures. As a result, the ethanol production of the evolved strain was increased by 22% in the presence of furfural. Moreover, the evolved strain showed an increase in glucose and xylose consumption rate by 36% and 10%, respectively. In addition, the strain was continuously cultured in the isopropanol containing medium for more than 140 subcultures. The ethanol production of the resulting strain was incressed by 70% in the presence of 30 g/L isopropanol. This strain also displayed an increase in glucose and xylose consumption by 50% and 30%, respectively.
