However, the effects and action mechanisms of [6]-shogaol on melanogenesis are not well known. This study aimed to evaluate the possible depigmentation mechanisms of [6]-shogaol. The results reveal that [6]-shogaol inhibits the expression of PAR-2, which control the deposition of melanosome in HaCaT keratinocytes. We also find that [6]-shogaol decreases the protein expression of tyrosinase-related protein 1 (TRP-1) and microhptalmia-associated transcriptional factor (MITF). The anti-melanogenic effect by [6]-shogaol is due to the activation of ERK signaling pathway. Moreover, MG-132 completely abolishes MITF downregulation by [6]-shogaol in B16F10 cells. This study concludes that [6]-shogaol stimulates the ERK pathway and subsequently induces MITF degradation, which leads to reduce melanin level and decrease melanogenesis.
