The medical science pays much attention to the combined therapy with Chinese and Western medicine gradually since developed, and focuses on medical plants. The aims of this study were to establish in vitro multiplication protocol of Taiwan native species, Petasites formosanus Kitam. and Dysosma pleiantha (Hance) Woodson. The processes include establishing aseptic explants,inducing shoots,rooting and transplanting. In the inducing shoots test, the test of Petasites formosanus Kitam. were BA 0.2 mg·L-1+NAA 0.02 mg·L-1,BA 0.4 mg·L-1+NAA 0.04 mg·L-1,BA 0.6 mg·L-1+NAA 0.06 mg·L-1,BA 0.8 mg·L-1+NAA 0.08 mg·L-1, with BA-NAA ratio 10:1 and the Murashige and Skoog (MS) medium was the control group.In vitro micropropagation of Petasites formosanus Kitam. was established to get maximum shoots multiplication rate by cultivating axillary buds on MS supplemented with BA 0.2 mg·L-1+ NAA 0.02 mg·L-1,but added plant growth regulators suppressing growth of explants and cause variation. In the rooting test, the test of Petasites formosanus Kitam. were MS, MS with 0.2 mg·L-1 IBA, MS with 0.5 mg·L-1 IBA ,1/2 MS, 1/2 MS with 0.2 mg·L-1 IBA, 1/2MS with 0.5 mg·L-1 IBA. Rooting rate achieved 100% by cultivating on MS plant medium for 2 weeks. 1/2MS with 0.5 mg·L-1 IBA has the most roots. Dysosma pleiantha (Hance) Woodson plant got the callus after being cultivated with different media and growth regulators, in which MS media with BA 2.0 mg·L-1+TDZ 0.2 mg·L-1induction of callus can continuously proliferate without browning and having died back, later generations to study. 廖松淵 2013 學位論文 ; thesis 40 zh-TW
