It is an exogenous retrovirus which has a worldwide distribution and infects domestic cats and, occasionally, wild cats. FeLV was discovered in early 1960s and can cause lymphoma and other fatal diseases. In this study, nucleotide sequences of long terminal repeats and nucleotide sequences of a core protein P 27 were compared and subjected into the phylogenetic analysis. Result showed that Taiwan strains clustered as a distinct clade than those strains from USA, Brazil and other countries. While sequencing Taiwan strains, a unique point mutation which was different from vaccine strains was presented on a core protein P 27. As the result, this facilitated an establishment of amplification refractory mutation system (ARMS) for the differential diagnosis of vaccine and Taiwan strains. An excellent specificity and sensitivity was proven for P 27 ARMS of FeLV. This system was further up-graded into the High Resolution Melting (HRM) platform. Not only a 100-fold sensitivity for the detection of virus than ARMS can be increased and all Taiwan strains, two vaccine strains, and mix strains can also be effectively distinguished.
