In this study, natural products isolated from Dioscorea opposite were subjected to study their protective effect against UVA or UVB irradiation in human dermal fibroblasts (HDF). These compounds were first analyzed for cytotoxicity and for their protective effect for HDF cell survival against UV irradiations. The effect on MMP-1 expression were then studied using wesern blot analysis. In addition, the effect on p38 kinase, ERK 1/2, and JNK signaling pathways and NFκB translocation were examined. Further analysis was performed to examine the effect of these compounds on the secreted MMP-1 level of HDF cells upon UV irradiation. Here we found that these compounds exhibited significant cytoprotective effect against UVA or UVB irradiation. In addition, these compounds also decreased the UVA- or UVB-induced MMP-1 expression and activation. In this study, two different natural products, M3 and M4, were identified and used for further studies on their signaling mechanism on UV-protective effect.. Our results indicated that the chemical constituents of Dioscorea opposite effectively inhibit MMP-1 expression and displayed potent anti-photodamaging efficacy in HDF cells.
