The major component of the polyol oils was found to be diacylglycerol, containing hydroxyl fatty acids. Thus it was to be designated as polyol DAG. It was found in preliminary functional test to likely possess an anti-inflammatory activity. Therefore, the aim of this study was to further characterize the anti-inflammatory effect of soybean polyols oil. Lipopolysaccharide (LPS)-induced inflammation in RAW264.7 macrophage was utilized as a model. Soybean polyol DAG significantly inhibited LPS-induced inducible expression of nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) as well as the production of nitric oxide and tumor necrosis factor-α (TNF-α). Characterization of the molecular mechanisms showed that polyol DAG supressed the activation of inhibitor of kappa-B kinase (IKK), the phosphorylation of inhibitor of kappa-B (IкB) and the nuclear translocation of factor kappa-B (NF-кB). However, polyol DAG did not inhibit LPS-induced activation of mitogen-activated protein kinases (MAPK) including extracellular signal-regulated kinase 1/2 (ERK 1/2), c-Jun N-terminal kinase (JNK), and p38. In animal test, soybean polyols DAG was confirmed to ameliorate skin inflammation in 12-O-tetradecanoylphorbol-13-acetate (TPA) – induced mouse ear edema. The inflammation process can be divided into the initiation phase and the resolution phase. In order to investigate whether soybean polyols DAG can promote the resolution of inflammation, the effect of soybean polyol DAG on the expression of some resolution-related factors, including transforming growth factor beta 1 (TGF-β1), 15-lipoxygenase (15-LOX), and 5-lipoxygenase (5-LOX) was analyzed. Consequently, polyol DAG inhibited the expression of 15-LOX and TGF-β1 but promoted that of 5-LOX . In summary, this study showed that soybean polyols oil possessed an anti-inflammatory activity and the underlying mechanism involved the inhibition of IKK/NF-кB pathway, but may not that of MAPKs. Whether soybean polyols oil promotes the resolution of inflammation need further investigation.
