Nervous necrosis virus (NNV) is one of the most devastating pathogens, infecting a variety of marine fish all over the world and bringing about high mortality in the larval grouper. Viral nervous necrosis (VNN) or viral encephalopathy and retinopathy (VER) is one of the most important viral diseases in aquaculture, and has caused mass mortality in more than 34 species of fish at larval and juvenile stages. Disease control would be an important issue for the world aquatic industry. Thus the prevention of vertical and horizontal NNV transmission has become critical. The immune system of grouper larvae is not mature enough to mount an immune response when vaccines are administered, so development of immersion and oral administration is necessary. In this study, we will use PCR to detect NNV. Then, we would use prokaryotic expression system to induce NNV coat proteins expression. After purification, protein was induced and the presence was confirmed by SDS-PAGE and Western Blot with anti-his antibody. In the future, protein with the formalin inactivated NNV to immunize laying hens, so that the eggs produced would contain immunoglobulins (IgY) specific for NNV. The IgY will be processed, isolation and purification. The goal of this study is to develop optimal passive immunity for protection against the most important viral disease of NNV in aquaculture.
