The objectives of this experiment are (1) to encapsule model drug (bovine serum albumin, BSA) by poly-alkylcyanoacrylate to avoid entero-degradation, (2) to improve the intestinal absorption rate by nanocapsule less than 200 nm, (3) to enhance humoral immune response by oral administrated Bacillus subtilis as adjuvant. The results showed: (1) the diameter of BSA poly-alkylcyanoacrylate nanocapsule is 80.82 ± 8.365 nm which is suitable for oral administration; (2) oral administrated BSA nanocapsule can induce BSA antibody titer increasing over time after each additional booster over 30 days period (p < 0.05); (3) BSA antibody titer is positively correlated to the dose of BSA nanocapsule (p < 0.05); (4) adding 5% B. subtilis as adjuvant can also enhance the humoral response (p < 0.05). Taken together, oral administrated BSA nanocapsule can effectively induce humoral immune response in grouper and provides the possibility for future development of oral vaccine technology.
