Summary:碩士 === 國立臺灣海洋大學 === 機械與機電工程學系 === 101 === Hemocompatility and adhesion of recombinant hirudin/collagen polyelectrolyte multilayers and heparin/collagen ones coated on a cp-Ti substrate via layer-by-layer self-assembly technique were investigated. These samples were pretreated by being immersed in 5M NaOH solution to obtain anionic surface with hydroxyl groups. The alkali-modified samples with a cationic surfaces were immersed in poly-L-lysine solutions. The repeated treatment of the samples were added by heparin (or recombinant hirudin) and collagan one, respectively. The surface topography, chemical composition, and hydrophilicity of the films were investigated by atomic force microscopy, scanning electron microscopy, Fourier transform infrared spectroscopy, and water contact angle measurement. The study of the adhesion of the multilayers was conducted by a nano-scratch test. The hemocompatility was evaluated by measuring the hemolysis ratio and platelet-covered area in vitro. The uncoated Ti surface and a previous developed collagen/heparin coated Ti sample were used as the benchmark. The results indicated that two kinds of anticoagulation multilayers succeeded coated on cp-Ti substrates. The increase of the multilayers’ thickness also enhanced their adhesion to the Ti substrate. The anticoagulation performance and the hemolysis ratio of recombinant hirudin/collagen multilayers on the titanium surface was not only superior to that of the uncoated Ti surface but also was even better than that of the collagen/heparin multilayer-coated Ti samples.
