The rate of aging and the appearance of age-related pathology are modulated by stress response and repair pathways that gradually decline. The objectives of this study were to investigate the effect of nucleic acid to brain function, immunity and reproduction performance in senescence-accelerated mouse prone-8 (SAMP8). Three and six month-old SAMP8 male and female mice were divided into control and 0.5% nucleic aicd (NC) group. After 12 weeks of feeding, body weight, food intake, aging score, open field activity test, active shuttle avoidance test and single-trial passive avoidance were performed during the experiment. The biochemical parameters of serum were analyzed after sacrificed. The activites of superoxide dismutase (SOD), catalase , glutathione peroxidase (GPx), malondialdehyde (MDA) and protein carbonyl concentrations in the liver and brain. The 8 - hydroxy-2’- deoxyguanosine (8-OHdG), nitric oxide(NO), nitric oxide synthase(iNOS), acetylcholinesterase (AchE) and pathological examination of brain. The proliferation of spleen cell and cytokines. The part two for 15 couples of 2 month-old SAMP8 to evaluation on litters and pups; motility , concentrations and abnormality of spermatozoa in male mice. The results showed that aging score significantly (p<0.05) decreased at NC group. NC group showed had significantly better learning and memory ability (p<0.05). The liver and brain SOD, catalase, GPx had significantly increase (p<0.05). The MDA and protein carbonyl concentrations in brain were significantly (p<0.05) decreased than control. The 8-OHdG, NO, iNOS, AchE and β-amyloid protein in brain were significantly (p<0.05) decreased than control. The proliferation of spleen cell, INF-γ and IL-2 were significantly (p<0.05) increased at NC group. In mice fed with NC group had better litters and pups. The sperm motility, sperm concentration in male NC group were significantly (p<0.05) increased than control group. The dietary nucleic acid supplementation has the potential to decrease oxidative street and brain β-amyloid improved the learning and memory, increased immunity and sperm quality of SAMP8.
