In the study One-factor-at-a-time experiments and Orthogonal Arrays was used to inquire Kaohsiung Mountain Cordyceps (C. takaomontana) into growth of optimal culture conditions of fruiting bodies on solid-state cultivation. Meanwhile, the effects of bioactive in different source of light and cultivation time with optimal culture conditions of fruiting bodies and substrate were discussed. By using the 95% ethanol or hot-water extraction to identify the differences of ability in the total polysaccharides, the total polyphenol content and DPPH radical scavenging were analyzed. The inhibition rate of anti-inflammatory, antioxidant and cancer cells was studied by using the cell function assessment. The results showed the optimum growth substrate of fruiting bodies was brown rice and the pH value was 5.5, the carbon source was molasses, the nitrogen source was Peptone. In hot-water extraction of total polyphenols, total polysaccharides, DPPH scavenging and determination of anti-inflammatory cells was better than 95% ethanol extraction. The source of light in the substrates of hot-water extraction was yellow and more content in total polysaccharide and total polyphenol. There was no significant difference in fruiting bodies. In cell function assessment, there was effectively inhibited A-549, Hep G2 growth and Nitrite generated and the antioxidant capacity was inefficient for 60 days solid-state cultivation.
