Tyzz-Yun-Gau was commonly being used as external using ointment since ancient of China, it mainly used on burns, sunburn, cuts, frostbite and ulcers, etc. It helps wound to recover and heal. Tyzz-Yun-Gau is a violet-red, applied to the skin easily stained in dress, causing inconvenience. If we can with patch, will improve this defect and more convenient to use. Tyzz-Yun-Gau was made traditionally with sesame oil, heat extraction Angelicae sinensis radix (ASR), and Arnebiae radix (AR). Because sesame oil can generate high temperature thus the effective ingredient of ASR and AR can be easily destroyed. Therefore, heat extraction by sesame oil is appropriate or not, should be studied. In order to fine the best extracted condition, we use four different solvents, including sesame oil, 50% ethanol, 95% ethanol, n-hexane, respectively, by ultrasound extraction and refluxation extraction to ASR and AR. TLC were the first to ferulic acid and shikonin marker compounds component qualitative analysis of ASR and AR,then were analyzed by HPLC, respectively. We choose the highest marker compounds solvents and methods for best extraction conditions. The results showed that the best extracted condition was by ultrasound with 50% ethanol extraction ASR and 95% ethanol extraction AR. The extraction condition also was used on extracting 1.25Kg of ASR and AR to make the Tyzz-Yun-Gau water-based patch, 10×14 cm, 12g per piece patch, and total had 810 pieces patches. In this study, we were established TLC to qualitative marker compounds and HPLC analysis validation to quantitative index compounds, ferulic acid and shikonin of Tyzz-Yun-Gau water-based patch. Ferulic acid and shikonin content were 347.6 and 11.7μg per patch, respectively. The results of this study will provide a reference of manufacture process and quality control of Tyzz-Yun-Gau water-based patch.
