Summary:碩士 === 國立中興大學 === 生命科學系所 === 102 === Malic enzyme (Malic Enzyme; ME) had three isoforms, respectively in the mitochondria, ME1 and in the cytoplasm, ME2 and ME3; which mitochondrial malic enzyme (Mitochondrial NAD (P) +--dependent malic enzyme; ME2) was major role in rapidly proliferating tissues, such as the spleen, thymus and tumor cells in which the catalytic substrates malate (Malate) thereby generating product pyruvate (Pyruvate), with NAD + or NADP + as a cofactor. ME2 catalytic reactions accompanied by NADH produced as the main source of energy metabolism molecule ATP, while in the fight against ROS damage may also play a very important role in helping to produce and promote NADPH Glutathione (GSH) maintained in a reduced state, effectively reduce the ROS injury. Because of ME2 metabolic pathways can produce NADH and procure GSH antioxidants remain in the reduced state, and accompanied by the generation of ATP, from above reasons to considers ME2 may have anti-apoptotic ability. To investigated weather ME2 overexpression prevented cells apoptosis, so my experiment study overexpressed ME2 in non-small lung cancer cells, H1299, A549 and acute lymphocytes leukemia cells, Jurkat then treated with H2O2, UV and curcumin. The AO staining and DNA fragmentation datas showed that ME2 overexpression cells more anti-apoptosis than ME2 vector, the ME2 overexpression cells might be through to the higher ability of ROS metabolic to anti-apoptotic. And in 2001 study had shown consistent associations between the cytoskeletal proteins -vimentin and cell apoptosis , the preliminary data had shown cell ME2 and vimentin had positive correlation. Further, we propose that?the anti-apoptosis ability of ME2 overexpression cell might be associated with the vimentin, and detail understand the mechanism of ME2 against cell apoptosis, hope can contribute for cancer treatment.
