Phylogeographic analysis of M. parviflora was conducted based on nucleotide variation of two chloroplast DNA non-coding regions (atpB- rbcL , trnL- trnF spacer) and internal transcribed spacers of nuclear ribosomal DNA from 100 individuals of 8 locations in Ryukyu Islands and Taiwan area. The cpDNA resulted showed, lower value of Fst indicated the population is moderately structured, and the variance occurred within populations. In constract, the nrDNA showed, significantly higher Fst value and suggested strong population differentiation between Ryukyu Islands and Taiwan area. The results of Minimum spanning networks analysis showed that both Taiwan and Ryukyu have diverse haplotype, but the variation of haplotype is higher in Taiwan for both shared and unique haplotypes . Therefore, we speculated that the population of M. parviflora in Taiwan possessed ancestral polymorphisms. In most populations, low nucleotide diversity, high haplotype diversity and negative value in neutrality test were observed. All the results indicated that the population of M. parviflora had been undergoing a rapid size expansion or bottleneck effect. The historical demography exploded was supported by mismatch distribution analysis and Bayesian skyline plot analysis. The study concludes that population size of M. parviflora might have increased through the continental bridge during the glacial period ca. 20,000~15,000. There is no significant due to the limited timespan for the evolution of this species. More samples from Guangdong and Ryukyu Islands, as well as more molecular markers would further elucidate the evolution history and phylogeography of M. parviflora.
