It might increase the morbidity of chronic metabolic diseases including type 2 diabetes, nonalcoholic fatty liver, hypertension, and cardiovascular diseases. The aim of this study was to investigate the effects of pure compound (1&apos;&apos;-acetoxychavicol acetate, ACA), essential oil (AGO) and ethanol extract (AGE) from Alpinina galanga (AG) rhizomes on 3T3-L1 preadipocytes differentiation and lipolysis in cell model. The results indicated that 15-125 μg/ml AGO could significantly inhibit the lipid accumulation by oil red O staining. The mRNA expressions of adipogenesis-related genes, including C/EBPβ, PPARγ, SREBP-1c, C/EBPα, GLUT4, LPL, CD36, aP2, ACC1 and FAS were significantly down–regulated during the 3T3-L1 preadipocyte differentiation. The heparin-releasable LPL activities were reduced by 31-125 μg/ml AGO, 7.8-31 μg/ml AGE and 0.75 μM ACA. Additionally, the lipolysis-related gene ATGL (adipocyte triglyceride lipase) and glycerol release were significantly increased by 31 μg/ml AGE. The anti-obesity effects of 2% LFA, 4% MFA and 10% HFA of Alpinia galanga freeze-dried powders (FA) were evaluated for 10 weeks on high fat diet (HFD)-induced obese BALB/c mice model. The results showed that the decreases of body weight and body fat ratio of LFA (8.7 and 2.3%), MFA (9.1 and 1.6%) and HFA(15.0 and 1.4%) groups were raised (p<0.05). Moreover, the sizes of epididymal fat cells were smaller in FA groups than HFD group. The serum parameters of triglyceride (TG), total cholesterol, aspartate aminotransferase (AST) and alanine aminotransferase (ALT) levels in LFA, MFA and HFA groups were significantly reduced which compared to HFD group (p<0.05). The histopathological analyses revealed that the lipid accumulation were attenuated and the scores of fatty liver were significantly decreased in LFA, MFA and HFA groups (3.9±0.3, 2.6±1.4 and 2.8±0.7) which compared to HFD group (4.8±0.4) (p<0.05). Administrations of FA could significantly promote the expression of lipid β-oxidation related-genes PPARα and CPT-1 up-regulation in the liver (p<0.05). In conclusion, the essential oil of Alpinia galanga can inhibit the adipogenesis pathway in 3T3-L1 preadipocytes and its freeze-dried powder potentially reduces the body weight, body fat ratio and severity of fatty liver in obese BALB/c mice by retarding the accumulation of fat. 蔣慎思 2014 學位論文 ; thesis 112 zh-TW
