Taiwan also have full life cycle aquaculture in several grouper species. However, the outbreak of Nervous Necrosis Virus infection severely affects the Taiwan grouper industry. In this study, we use several Chinese herbs as immune stimulants to treat grouper cell. By using MTT assay and real-time RT-PCR, we evaluate the antiviral activity in grouper cell. Furthermore we also try to exhibit the antiviral mechanism in these Chinese herbs. In the extract of Polygonum cuspidatum, Astragalus membranaceus, Houttuynia cordata treatment group, the antiviral activity is higher than control group by dose response. However, the extract of E. sinica and Bupleurum spp. cannot observe significant difference between treatment and control group. The similar observation was also being shown in virus titer assay. By analyzing the immune gene expression, we found that these Chinese herbs could regulate the innate and adaptive immunity such as IL-6, IL-1β, TNF-α, Mx, C3, MHC-1α and IgM.
