The aim of this study was to investigate the preventive effects of fat accumulation by Curcuma phaeocaulis and Curcuma aromatica in vitro and in vivo. The in vitro results indicated that 50-100 g/ml CPO and 500g/ml CAE can significantly inhibit the lipid accumulation by oil O staining. RT-PCR (real-time polymerase chain reaction) analysis was used to estimate the gene expression of adipocytes differentiation and lipid metabolism. The results showed that 100 g/ml CPO treatment can decrease the expression of adipogenesis-related gene ACC (53.4±12.4%) and increase the expression of lipolysis-related gene CPT-1 (91.0±52.2%). 500g/ml CAE treatment can decrease the expressions of preadipocytes differentiation-related gene PPAR(54.5±14.5%) and ACC (67.4±8.8%), but increase the expression of CPT-1 (168.6±57.7%). The preventive effect of fat accumulation by Curcuma phaeocaulis and Curcuma aromatica powder (CPP and CAP) was evaluated in vivo. Eight-week-old male C57BL/6J mice were fed normal diet as control group (n=10) and high-fat diet to induce obesity (n=70) as obese group for ten weeks. Subsequently, obese group mice were randomly divided into seven groups (n=10), including high-fat diet group (HFD); low, medium and high-doses of Curcuma phaeocaulis group (LCP、MCP and HCP) and low, medium and high-doses of Curcuma aromatica group (LCA、MCA and HCA). These mice were fed for eight weeks and sacrificed, the serum biochemical parameters and histopathological analysis were assayed. The results showed that administration of CA and CP powders reduced the ratio of body weights range from 19.4-28.8 % and 14.4-36.5 % (p<0.05), and body fat ratio from 1.6-3.1 % and 0.4-4.0 %. Moreover, the size of adipocytes was smaller in all treatment groups than HFD group. The serum parameters of AST, ALT and leptin levels in treatment groups were reduced compared to HFD group. Histopathological analysis revealed that LCP, MCP, HCP, LCA, MCA and HCA decreased the pathological change of fatty infiltration in liver from score 4.5±0.8 (HFD group) to score 2.6±1.6, 3.1±1.2, 2.1±1.3, 3.8±1.0, 3.4±1.4 and 1.3±1.3, respectively. In conclusion, essential oil of Curcuma phaeocaulis and ethanol extract of Curcuma aromatica can inhibit lipid accumulation in 3T3-L1 preadipocytes by decreasing the expression of ACC and increasing the expression of CPT gene. Furthermore, oral administration of CP and CA powder can reduce the body weight, body fat ratio, fat accumulation in epididymal fat and severity of fatty liver in the mice.
