As the population who suffer from allergy is rapidly growing, fast and accurate detection of allergy becomes highly demanded. At present, the main method to diagnose allergy is to detect immunoglobulin E (IgE) or to examine internal skin to know how allergy develops. However, not all allergic reaction is related to IgE because the main reason leading to allergy is due to basophils or mast cell, which secretes the allergic composition such as histamine after getting activated or stimulated. As a result, having a test on the allergic reaction material is seen to be direct and important. This experimental study aims to observe the properties and exocytosis of single basophil cells trapped on a microfluidic platform equipped with impedance measurements. The lithography technology was used to manufacture microfluidic platform composed of microchannel structure and microelectrodes chip. The FEM software COMSOL is employed to simulate the flow and electric fields. The microfluidics designed utilizing the hydrodynamics filtration technique is able to capture single cells in all of the 20 trapping chambers. The impedance between the microelectrodes fabricated on the trapping chamber was measured for a frequency range of 100 Hz to 100 kHz at a supply voltage of 0.5V. The impedance measurements show significant increase in magnitude and change in phase for the cases of trapped single basophil cells. The single basophil stimulated by calcium ionophone was found to largely decease in magnitude of the impedance measured in 15-30 min. The measurements are further used to build an equivalent circuit for calculation of the cell resistance and capacitance.
