Summary:碩士 === 國立彰化師範大學 === 生物技術研究所 === 103 === OTA is a virose fungal secondary metabolites, mainly produced by certain Aspergillus and Pencillium strains, which can contaminate food, vegetables, coffee, grapes cocoa and other cash crops. It has been widespread around the world, while it could contaminate a large number of species, and the producers can adapt to a extensive environment. It has a strong renal toxicity, liver toxicity, teratogenicity, immune toxicity and embryo toxicity. Many countries have established limits, so the establishment of low cost, fast and efficient field detection is extremely important. Antibody test are widely popular due to its rapid, high sensitivity, high specificity, portability and easy operation. But the monoclonal or polyclonal antibody isolated from immunized animals is a time-consuming experiment. In our study, we are able to quickly and effectively to obtain specific antibodies instead of isolating from the immunized animals. Therefore, we use a computer-aided antibody design to imitate the natural effective properties of antibodies such as variability of amino acids and specific length of CDR, which are employed to design specific primers for phage display antibody libraries. Phage display has been developed as a robust technology offering great potential for design of synthetic antibody libraries. So far, we obtained two specific antibodies against OTA antigen. In the future, two antibodies will be employed to carry out affinity maturation for generating high-affinity antibodies against OTA antigens.
