Current eel culture depends entirely on glass eels captured from the wild. However, eel populations have declined in recent years and recent efforts have not yet succeeded in the mass artificial production of glass eels. The decrease in the production of the glass eels leads the increase of the price. Thus, other species of eels were passed off for Anguilla japonica. Mitochondrial DNA is widely used in genetic analysis, phylogenetic analysis, biological diversity and species identification. In this study, two mitochondrial genes, 16S rRNA and COI, were used to distinguish the eel species. We established a rapid, inexpensive and reliable method by polymerase chain reaction. For four eel species, primers were designed and tested to distinguish the eel species. The results showed that the specific primers could identify eel species successfully.
