We attempted to find the whitening activity compounds present in many herbal medicines used for this purpose we also discuss their mechanism in melanin biosynthesis. In this study, we investigated the effects of Ligustrum lucidus Ait extract on melanogenesis, including the activation of melanogenesis signaling pathways. This results suggest that Ligustrum lucidus Ait extract inhibited include melanin synthesis and tyrosinase activity and melanogenesis-related proteins such as microphthalmia-associated transcription factor. In addition, Ligustrum lucidus Ait extract not only acts by phosphorylating but also activating melanogenesis inhibitory proteins such as Akt and mitogen-activated protein kinase (MEK)/extracellular signal-regulated kinase (ERK). These results suggest that activation of the MEK/ERK and PI3K/Akt signaling pathways is involved in the melanogenesis signaling cascade, and that activation by Ligustrum lucidus Ait extract reduces melanin synthesis via down-regulation of MITF and its downstream signaling pathway.
