The aim of this study is to investigate the expression of the sodium potassium ATPase (NKA), a transmembrane protein consisted of a pair of α- and β-subunits for transport of Na+ and K+, in epithelium of gastric adenocarcinoma as well as the potential roles of NKA in cell homeostasis and adherens junction. The data are expected to be used as the bases to establish the NKA as a biomarker and to develop the screening method. The results of immunostaining revealed that the NKA α- and β-subunits were colocalized with E-cadherin (adherens protein) in the gland of gastric epithelium. Both NKA activity and α1-isoform protein expression were reduced in the study group, implying the impairment of NKA functions. In the adheren junctions, NKA β1-isoform and E-cadherin also showed a significant decrease in the study groups, indicating the adhesion force in-between cells might be weaken. In addition, the results of dot blotting were similar to those of immunoblotting, confirming the significant differences of various proteins between the study and control groups. Taken together, significant changes in NKA expression (protein and activity) in tumor regions showed a promising biomarker for gastric adenocarcinoma. Therefore, developing certain screening methods for detecting NKA expression (protein/activity), such as dot blotting and others, may be useful in early diagnosis of gastric adenocarcinoma. 李宗翰 李建達 2016 學位論文 ; thesis 59 en_US
