In this study, we investigate the effects of different fertilizers, zinc application and planting media on the growth and accumulation of active ingredients of agarwood (Aquilaria agallocha Roxb) seedling. High performance liquid chromatography (HPLC) and UV-Vis spectrophotometer were used to detect the content of active compounds, such as mangiferin, rutin, quercetin, epigallocatechin gallate (EGCG), total phenolics and total flavonoids. With high level organic fertilizers, it can promote the growth of agarwood seedling, but no significant effect of Zn application on the seedling growth. It exhibits highest growth and accumulation of mangiferin and rutin under akadama treatment among planting media study. On the other hand, the annual variations of active compounds in leaves of 20~25-year-old agarwood tree in field have been analyzed. Following the manufacturing processes of green tea and black tea, we have investigated possible variations of active ingredients and antioxidative activities of agarwood leaf tea in order to explore the optimal harvest time and the difference of their health functions. It reveals that the content of mangiferin-main active compound of agarwood leaf fluctuates annually. It reaches highest amount in July with 53.31 mg g-1 DW, circa 4.94 fold higher than the lowest amount obtained in February. For comparing experiments by analyzing the content of active compounds of agarwood leaf tea with manufacturing processes of green tea and black tea, it shows that the content of quercetin and EGCG will change under different fermentation temperature and the best fermentation condition is at 28℃ for 6~8 hr. But the content of quercetin can not be detected under the fermentation treatment at 33℃ for 4 hr. Agarwood leaf tea would be a highly potential herbal tea. It will exhibit its optimal health functions with the most suitable harvest time and manufacturing process. This results suggest that the best agarwood tea will be obtained when harvested in July at higher ambient temperature and under manufacturing process of green tea.
