Recent advances in luminescence thermometry have made it possible to detect the transient temperature change with high spatial resolution and sensitivity using fluorescent nanodiamond1. Here, we show that gold nanorod-fluorescent nanodiamond (GNR-FND) hybrid is produced as a two-in-one (heater & thermometer) nanoprobe to probe the local temperature and thermal structural stability. Polyarginine-coated FNDs were mixed with citrate-capped gold nanorods (GNRs) to form a dual-functional nanoprobe via multivalent electrostatic interactions. With the use of a 594 nm laser for both heating and probing, we are able to determine the transient temperature jumps by recording the spectral shifts of the zero-phonon lines of negatively charged nitrogen-vacancy centers in FNDs. A significant temperature jump (45±2K) induced by using differential Laser-induced heating on single GNR-FND was observed. The technique allowed us to quantitatively characterize the transition temperature (Tm) at which the intercellular membrane nanotube could break down. Our results demonstrates the capability of GNR-FND hybrid for efficient local heating and temperature sensing, which paves the way towards in vivo characterization of local thermal stability of cellular structures in all living things.
