C. inophyllum leaf has been used as a traditional medicine to treat traumatic bleeding, bruises, rheumatism, etc. Chemical composition of the leaves, stems, root bark, seeds and other parts of the C. inophyllum includes xanthones, flavonoids, terpenoids, coumarins and others. Objective: This study was to investigate the effects of different pretreatment methods on the oil production and composition change of seed oil and seed kernel extract of C. inophyllum. Materials and Methods: The mature seeds of C. inophyllum were collected from the Ao-Drum Wetlands of Chiayi, Taiwan. After pretreatments of the seeds by natural drying, artificial drying, freeze drying, hexane and ethanol were used sequentially and traditional pressure preparation was also applied to extract oils. The differences in oil production and their compositions were analyzed by Spectroscopy (ATR-FTIR, UV-vis) and chromatography (GC-FID, HPLC). Results and Discussion: C. inophyllum seed oil contained 29.05~30.90% saturated fatty acids (C16:0 and C18:0) and 66.93~69.34% unsaturated fatty acids (C18:1 and C18:2), which were lower than commercially olive oil (80.63%) and camellia seed oil (85.85%). Pretreatment methods changed the yield and components in oil. The amount of saturated fatty acids and oleic acids were the highest in artificially dried oil than the oils from naturally drying and freezing process. On the contrary, the latter two contained more unsaturated fatty acids than the former. Moreover, the oil from pressed cake contained the highest amount of oleic acids and unsaturated fatty acids and the lowest amount of saturated fatty acids. Indeed, pretreatments could change oil yield. All the oils from different pretreatments possessed characteristic absorption peaks at 2925 cm-1, 2854 cm-1,1746 cm-1, etc for fatty acids determined by FTIR. UV absorption spectrum revealed that C. inophyllum seed oil also possessed characteristic absorption peaks of coumarin at 274 nm, 280 nm and 313 nm. Furthermore, HPLC analysis revealed two absorption peaks at 313 and 280 nm, indicating the possible presence of coumarin.
