Numerous plants are thought to have a potential value for working as antioxidant and anti-inflammatory agents through quenching free radicals, increasing antioxidant defenses, or inhibiting the release of proinflammatory mediators. Glechoma hederacea belongs to the Labiatae family and has many biological effects. The objectives of this study were to evaluate anti-oxidatant and anti-inflammatory activity of HWG. The result of in vitro study had shown that the HWG possessed anti-oxidative characterstics including α,α-diphenyl-β-picrylhydrazyl, 2,2’-Azino-bis (3-ethylbenz-thiazoline-6-sulfonicacid) radical-scavenging effects,reducing power and lipid peroxidation-inhibitory effects. It was also found that anti-oxidatant activities of the HWG increased with increasing concentractions. Furthermore, our results indicated that treatment of RAW264.7 cells with the HWG significantly decreased LPS-induced nitric oxide (NO) content, superoxide anion levels, reduced thiobarbituric acid-reactive substances (TBARS), increased GSH levels and regulated the antioxidant enzyme activities. The results showed that the HWG may ameliorate oxidative stree in LPS-induced RAW264.7 cells. Moreover, the HWG can prevents LPS-induced DNA damage in RAW264.7 cells, reduce the LPS-induced RAW264.7 cells mRNA expression including iNOS, COX-2, TNF-α, IL-6, IL-1β and IFN-β, and reduce the LPS-induced RAW264.7 cells protein expression including iNOS, COX-2 and TLR4.
