Summary:碩士 === 靜宜大學 === 食品營養學系 === 104 === Essence of chicken (EOC) might reduce the damage of reaction oxygen species (ROS) by the capacity of antioxidantive component such as carnosine and anserine. The experiments use 3-month-old SAMP8 male and female mice were used in this study. SAMP 8 mice were randomly divided into four groups, where each group contained 15 mice, control group (1.00ml double-distilled water) and three experimental groups supplemented with 7 ml/kg BW/day, 14 ml/kg BW, and 28ml/kg BW Essence of chicken for 12 weeks. The body weight, food intake and water consumption were measured every week. The 12th and 13thweek, the open field activity test was recorded in evaluation for behavior. One week before scarification, the single-trail passive avoidance test and active shuttle avoidance test were performed. The biochemical parameters of serum were analyzed and, thiobarbituric acid-reactive substances (TBARS) superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) and catalase (CAT) activities of liver and β-amyloid protein (Aβ) and thiobarbituric acid-reactive substances (TBARS) and 8-hydroxydeoxyguanosine (8-OHdG) of brain after the mice sacrificed. The results showed that experimental groups, mice in body weight, food intake and water intake,activity levels and organ weights in both control group and the experimental groups have no significant differences. The aging score of experimental groups were significantly lower than the control group (p<0.05). Results from single-trail passive avoidance test and active shuttle avoidance test showed that experimental groups had significantly better learning memory ability (p<0.05). The TBARS of liver in the experimental groups were lower than control groups (p<0.05). Activites of antioxidative ability, including SOD, GPx and catalase , experimental groups were significantly higher than that of control group (p<0.05). The biochemical parameters of serum were triglycerides and total cholesterol, experimental groups has better significantly difference (p<0.05) compared with other groups.Administration of EOC significantly decreased the level of 8-OHdG, TBARS and β-amyloid protein deposition in the brain (p<0.05). In summary, the supplements of EOC could improve the learning-memory ability and antioxidative defense system and the accumulation of β-amyloid protein, promote the antioxidative defense system, and refarded the aging process.
