Many natural products with anti-cancer properties exist in the environment. Several studies have shown that Lonicera japonica Thunb. has anti-cancer and anti-inflammatory properties; its luteolin shows an anti-cancer effect and its chlorogenic acid shows an anti-inflammatory effect. Thus, the aim of this study was to investigate the effects of L. japonica Thunb. administration on the mechanism of apoptosis and anti-inflammation in the human colon cancer cell line HT-29 and anti-inflammation in ICR mice. The cell viability ( Cell Viability ) assay showed that an aqueous extract of L. japonica Thunb. ( LjWE ) showed significant time and dose-dependent inhibitory effect on the HT-29 cells. A clear DNA ladder-like pattern appeared after 60 h of LjWE treatment. Flow cytometry analysis revealed that HT-29 cells were significantly arrested in the G0/G1 phase after 24, 48, and 60 h of treatment with 500 μg/ml of LjWE Western blot analysis showed that treatment with 500 μg/ml of LjWE resulted in increased expression of p53, caspase-9, caspase-3, and Bax, which further inhibited the amount of Bcl-2, and reduced the expression of inflammatory factors such as NF-κB, TNF-α, IL-8, and IL-6. In addition, a mouse-ear swelling model in an animal study revealed that an LjWE could inhibit skin inflammation. Overall, these results showed that the LjWE could promote apoptosis of HT-29 cells by increasing the expression of p53, caspase-9, and caspase-3, and could inhibit inflammation by reducing the expression of NF-κB, TNF-α, IL-8, and IL-6.
