Description
Summary:碩士 === 大同大學 === 生物工程學系(所) === 104 === The main technique of the present study is using polymer film which made with biodegradable materials polycaprolactone (PCL) by electrospinning technique. The polymer film without colchicine was used as the blank and which containing colchicine polymer film was used for the study. The polymer film was cut into the appropriate size to fit 24well plate, on which NIH-3T3 mouse fibroblasts was cultured to investigate the release of colchicine from PCL fiber membrane. After 4, 24, 72 and 120 h of incubation, effects of medicated and non-medicated on the growth of cell membranes were observed. After the first four hours of incubation, maximum release of colchicine was achieved and this was sustained up to 48 h. The effect of drug released from the film on inhibition of NIH-3T3 mouse fibroblast growth was determined by MTT assay. The results showed that the film carring the drug effectively inhibit the growth of fibroblasts within 24 h. After 36 h fibroblast cells could grow normally again, because no more drug was released from the film. In conclusion, colchicine treated PCL electrospun fibers can effectively inhibit the growth of NIH-3T3 mouse fibroblast cells.
