Background:The incidence rate of diabetic peripheral neuropathy accounts for approximately 50-60% of all patients with diabetic mellitus. The entire mechanism of diabetic peripheral neuropathy remains unclear. Besides, the efficacy of treatment toward patients with diabetic peripheral neuropathy is still dissatisfied to clinical physicians currently. Our study majorly aims to evaluate the therapeutic effect of different dosage of “Huangqi Wuwu Decoction (also named Huangqi Guizhi Wuwu Decoction)” in treating diabetic peripheral neuropathy among neuropathic model in diabetic rodent models. Methods:We choose male Sprague-Dawley rats, weighing between 200-250 g to apply for the animal models. Then, we use the method intra-peritoneal injection with streptozotocin (STZ) solution to induce hyperglycemic state. After confirming the diabetic Sprague-Dawley rats models been established by checking the value of fasting glucose, we observe the rats’ behavior and speculate diabetic peripheral neuropathy being happening gradually among radiant heat test and rotarod test. We once again manipulate the radiant heat test, rotarod test, measuring nerve conductive distal latency and amplitude, observing the biopsy of gastrocnemius muscle and sciatic nerve, calculating the density (axon numbers of sciatic nerve in certain area) and doing immunohistochemistry stain after treating three different dosages (0.5 g/Kg, 1 g/Kg, 2 g/Kg) of “Huangqi Guizhi Wuwu Decoction” solution to diabetic Sprague-Dawley rats models. In addition, we further evaluate the results by using statistical software. Results:Especially in low dose (0.5 g/Kg) intervention of “Huangqi Guizhi Wuwu Decoction” solution in experiment group toward the male Sprague-Dawley rats in hyperglycemic state, the p value calculated by statistical software was 0.0081 in rotarod test and 0.0129 in density measurement (axon numbers of certain area) of sciatic nerve, both less than 0.05, and reached statistical significance. The p value was 0.0776, close to 0.05 in nerve conductive distal latency measurement. The biopsies of gastrocnemius muscle and sciatic nerve also have shown relative findings and characteristics of diabetic peripheral neuropathy among the diabetic Sprague-Dawley rats. However, we couldn’t refuse the null hypothesis in comparing nerve conductive amplitude due to the fact that the p value was 0.3346, more than 0.05. Conclusions:Despite “Huangqi Guizhi Wuwu Decoction” solution could not change the hyperglycemic state induced by diabetes mellitus, yet the diabetic Sprague-Dawley male rats under hyperglycemic state treated in low dosage (0.5 g/Kg) indeed appeared much better in motor coordination, nerve conductive distal latency and unit density measurement. On the other hand, the aspects of increasing total numbers of diabetic Sprague-Dawley male rats, prolonging experimental course duration, and the safety of long-term taking “Huangqi Guizhi Wuwu Decoction” solution could be investigated and verified in further studies in the future.
