It has several biological including anti-inflammatory, anti-tumor, antidiabetic, antibacterial, antifungal, antiparasitic, antioxidant and anti-obesity activities. The purpose of this study was to investigate the melanogenesis inhibitory properties and antioxidant properties of α-mangostin. Assays of mushroom and intracellular melanin contents and tyrosinase activites were conducted spectrophotometrically while expression profiles of the melanogenesis -related proteins were determined by Western blotting. Further, The possible signaling pathways involved in α-mangostin-mediated inhibition of melangenesis were also survey from specific protein kinase inhibitors. The result from the study indicated that α-mangostin (30-300μM) inhibits mushroom tyrosinase activity in a dose-dependent pattern. The, α-Mangostin repressed α-MSH induced intracellular tyrosinase activity and reduced melanin content in mouse B16F10 melanoma cells . α-Mangostin also suppressed the intracellular reactive oxygen species (ROS) level in a dose dependent matter. The results showed that α-Mangostin inhibited melanogenesis in B16F10 cells via activating PI3K/AKT (Protein kinase B) pathways. And reducing the level of intracellular ROS.
