The microfluidic chip is fabricated using a CO2 laser system and comprises a serpentine channel, a mixing chamber, a sample chamber, two reagent chambers, a reactive chamber, and three rectifiers based on PDMS membrane. The PDMS membrane deflects in the upward direction during forward flow; thereby permitting the fluid to flow over the obstacle. The main components of the small operated system include a power source, a relay, a microfluidic chip holder, a barometric value, and a temperature controller module. In the proposed device, the KHV sample and a LAMP reagent are loaded into the chambers and an external gas pressure driving force is then applied to mix the sample and reagent. Meanwhile, a constant temperature of approximately 60°C is kept for approximately 60 min in order to replicate KHV sample by the small operated system. Finally, the replication KHV sample and SYBR green reagent are mixed to detect the KHV using fluorescence detection method. A gel electrophoresis method is confirmed proposed integrated microfluidic platform. Overall, the results presented in this study show that the proposed integrated microfluidic platform provides a compact and reliable tool for replication KHV sample and measurement purposes.
