Recently, liver cell transplantation has become an alternative way to liver transplantation, can be used to treat liver disease. However, previous study found that cells may easy to fall directly into the bottom when culturing in the porous scaffold so that cells are not easy to stack and grow. This study designs a new scaffold to compare with another scaffold which used in the past experiment and through the way of dyeing to enhance the accuracy of scaffold. Hope to improve cell viability and proliferation. The main materials are PCL-DA and PGSA. The effects of different crosslinking densities were compared by tensile tests, contact angle tests and thermal properties analysis. Add the Sudan Black to improve the accuracy. At the concentration of 0.04wt%, the accuracy error can be controlled at 1~2%. After the results of the above-mentioned test, PGSA60 + PCL-DA was used to fabricate scaffold and the hepatocytes culture. It was found that the step-type scaffold has better cell viability due to it has more area which the cells could attach. Yih-Lin Cheng 鄭逸琳 2017 學位論文 ; thesis 103 zh-TW |
