It is well known that fucoidan have immunomodulatory and anti-cancer effects. However, the chemical components of fucoidan and the anti-tumor ability of fucoidan isolated from different seaweed species were unclear. In this study, we found that fucoidan extracted from different species of brown seaweeds have significantly different components and the anti-non-small-cell lung cancer (NSCLC) (A549) ability. Comparisons were made among Fucus vesiculosus, Laminaria japonica and Sargassum hemiphyllum. The fucoidan extracted from F. vesiculosus was the most one types in anti-NSCLC. In addition, the specific fucoidan extracted from Laminaria japonica and Sargassum hemiphyllum have a high levels of free form sugars, such as sorbitol and mannitol. Owing to overtake sorbitol may cause diarrhea and other side effect, we use four ways to remove the free form sugars of fucoidan and examine whether the anti-cancer effect can be improved. The results showed that the removement of the free form sugars could only improve the anti-tumor efficiency by a small percentage. Dialysis method could be a simple and rapid way to remove the free form sugars of fucoidan. Furthermore, we investigated the fucoidan-induced transforming growth factor receptor (TGFR) degradation and apoptosis were suppressed by knockdown-Tollip/TLR4. In summary, different resources of fucoidan have different effects on anti-tumor ability and using purified fucoidan could prevent patients ingested large amount of sorbitol and avoid the possible side effect.
