| Summary: | 碩士 === 國立臺灣海洋大學 === 水產養殖學系 === 106 === Nervous necrosis virus (NNV) is a kind of betanodaviruses, which causes viral nervous necrosis (VNN) and it has been the root cause of great economic loss for aquaculture industries. Grouper is an important aquaculture species with high economic value in the Asia Pacific region. They are susceptible to piscine nodavirus infection during young stage, which results in viral nervous necrosis disease that causes 100% mortality. In this study, we stimulate the innate immune responses in vivo and in vitro as prevention of the NNV invasion by using Laminariales japonica fermenter extract. In vivo experiment, the Laminariales japonica fermenter extract was grounded into powder and mixed into the feed as an immune stimulant for a week. After that, we analyze the immune gene expression (IL-6, IL-1β, TNF-α, Mx, C3, IgD, IgM, MHC I and MHC II) of each group. Besides, in vitro experiment, we used different concentration of Laminariales japonica fermenter extract as immune-stimulant to treat grouper head kidney cell. We used MTT assay to calculate cell viability and real-time PCR to determine the expression of immune genes. In our results, cold extracts of Laminariales japonica fermenter may not induce the mRNA expression of immune genes in groupers after NNV infection; however, 10 mg/g cold extracts of Laminariales japonica fermenter would partially inhibit the gene expression of NNV. Therefore, we considered cold extracts of Laminariales japonica fermenter may have the ability against NNV. Laminariales japonica fermenter extract may have potential as an immune stimulant to replace vaccine.
