In recent years, jabuticaba become an attractive fruit in Taiwan, because it is rich in vitamins, minerals, carbohydrates, and antioxidants. Previous studies have reported that different biologically active compounds from jabuticabas can be used to prevent or diminish various diseases such as cardiovascular and cancer. In this study, the extracts from jabuticabas’s peels and seeds were extracted with different extraction methods for biological activity. The extraction conditions such as water extraction, ethanol extraction (50 % and 95 %), shaking extraction, hot water extraction and sonication extraction have been conducted during this study. In order to explore the bioactivities of peels or seeds from jabuticabas, antioxidant activity assay (DPPH scavenging free radical, chelating ferrous iron, total polyphenol content detection and DCFH-DA on flow cytometer), MTT assay, cell cycle analysis and gelatin zymorgraphy assay (MMP-2 and MMP-9) have been investigated. The results have shown that the antioxidant activity, the extracts of water extraction from jobuticaba’s peels and seeds, has better anti-oxidation ability compared with ethanol extracts, however, the antioxidant ability of the seed extracts is better peel’s extracts. The results, from MTT assay, have shown that the extracts from jabuticaba’s peel and seed can significantly inhibit the cell growth of breast carcinoma cell (MDA-MB-231). According to western blot and flowcytometry analyses, the results have also shown that the jabuticaba’s extracts can significantly increase the oxidative stress on MDA-MB-231 cells and cause these cells arrested at Sub G1 and G2/M phases. According to the gelatin zymorgraphy assay, the peel’s and seed’s extracts from jabuticabas can significantly inhibit the protein secretion of MMP-2 and MMP-9 on human fibrosarcoma cells (HT-1080). Therefore, the results shown as above, the jabuticaba’s peels and seeds have highly potential to develop as functional foods. CHEN,JIAN-CHYI 陳健祺 2018 學位論文 ; thesis 82 zh-TW
